You need only add template and primers, reducing set-up time by half. It is effective for either short or long targets. PCR SuperMix, High Fidelity, contains Mg⁺⁺, dNTPs, and a mixture of recombinant Taq DNA polymerase and the proofreading DNA Polymerase from Pyrococus species GB-D, at concentrations sufficient to allow amplification during PCR. You will produce products that are mixed blunt and 3-A' ends; however, the majority of ends will have 3-A' overhangs.Using PCR SuperMix, High FidelityPCR SuperMix, High Fidelity, is supplied at a 1.1X concentration to allow approximately 10% of the final reaction volume to be used for the addition of primer and template solutions. This ready-to-use reagent offers a 6-fold improvement in fidelity over Taq DNA Polymerase and is effective in amplification over a large range of target sizes. Targets beyond 12 kb in length may require optimization of PCR cycling conditions and success is heavily dependent on primer and template quality.
