Amyloid beta (A4) precursor protein (APP) is a 100-140 kDa transmembrane glycoprotein existing as several isoforms. The amino acid sequence of APP contains the amyloid domain Abeta (Aβ), which can be cleaved by a two-step proteolytic cleavage and release Aβ peptides of Aβ [1-40], Aβ [1-42], and Aβ [1-43] corresponding to residues 40, 42, and 43 respectively. The extracellular plaque deposition and accumulation of the released Aβ peptides (β -amyloid), and intracellular neurofibrillary tangles (NFTs) in the brain, are the characteristics of Alzeimer disesease (AD). Increased release of the ‘longer forms’ of Ab peptide, Ab42 or Ab43, which have a greater tendency to aggregate than Ab40, occurs in individuals expressing certain genetic mutations or expressing certain ApoE alleles, or may involve other, still undiscovered factors. It has been shown that this increased release of Ab42/Ab43 leads to the abnormal deposition of Ab and the associated neurotoxicity in the brains of affected individuals.
