CD154 (Anti-Human CD40L, gp39, CD40 ligand) recognizes a 39-kDa antigen that is a member of the tumor necrosis factor (TNF) receptor family. CD40L is transiently expressed on activated CD4 T lymphocytes. The expression of CD40L by activated T-helper cells triggers B-cell cycling through binding to CD40. This co-receptor interaction is required for B-lympho-cyte maturation, response to T-dependent ligands, and isotype switching. T-lymphocyte proliferation and generation of cytotoxic T lymphocytes (CTLs) is associated with the presence of CD40L. Defects in the CD40L gene are associated with the X-linked hyper IgM syndrome.CD154 antigen is found predominantly on activated CD4⁺ peripheral T lymphocytes, as well as on a small fraction of CD8⁺ lymphocytes, certain malignant cells, mast cells, and basophils. Mature CD4⁺CD8^- thymocytes also express CD40L following stimulation.
