Specifically, Mena is involved in cell polarity, cell motility, cytoskeleton remodeling, filopodial, and lamellipodial dynamics within migrating cells. Three forms of Mena are expressed in embryonic head tissues and are identified based on molecular weight (140 kDa, 88 kDa, and 80 kDa). In adult wild-type tissues, Mena has been isolated in brain, testis, ovary, and fat in the 80 kDa form. Studies show the 140 kDa form is expressed only in adult brains while the 88 kDa form decreases to undetectable levels by E16, (Lanier, 1999). Mena contains 4 domains of interest; the Ena/VASP homologue 1 (EVH1) domain, an EVH2 domain, a proline-rich domain, and an Arg/Leu/Glu-risch region. Expression of Mena has been observed in the myoepithelia of breast, bronchial glands, parotid, and sweat glands. Mena has also been observed in the epithelium of mouse mucosal tissues: colon-rectum, endometrium, pancreas acinar ductal, placenta vascular media, and prostate fibromuscular stroma. Required for the actin-based mobility of Listeria monocytogenes (by similarity). SUBUNIT STRUCTURE: Homotetramer by similarity. Interacts with APBB1IP, APBB1, PFN1 and ROBO4. Isoforms, containing the polyproline-rich regions with PPLP motifs, bind the WW domain of APBB1IP. Isoforms, containing the PPSY motif, bind, in vitro, to the WW2 and WW3 domains of NEDD4 and to the WW1 domain of YAP1.
