    View All» Application Notes: Immunohistochemistry on paraffin embedded tissue: 5 μg/mL, with microwave antigen recovery. The method used for these antibodies is found in Metzer et al 1999, Kidney Int 56:1442-1454. Briefly, 2-4μm sections were cut followed by overnight drying in 37C. Dewaxed in xylene for 10min, followed by 10 in acetone and 10 min acetone/TBS 1:1. After this treatment the slides were washed in TBS, placed in microwave proofed tubes containing 0.01 citrate buffered solution pH7.0 and were boiled for 5 minutes 600W in the microwave. Evaporated volume was replaced with distilled water and the procedure repeated four times. After microwave treatment the slides were left to cool down and washed with TBS. (Control frozen sections were fixed in acetone 10 minutes and air-dried). The researchers used an ABC or APAAP enhancement to detect ACE. Other antibodies to ACE has been used successfully for: 1) the study of ACE expression during human pathology (myocardial infarction, pulmonary hypertension, atherosclerosis, kidney diseases Balyasnikova, Franke, Pauls, Metzer & Chenard-Neu references) as well as using animal models (Franke,Morrell 1997, Haberboshc references), 2) testicular cancer diagnostics (Falkenhahn, Morrel, 1995) and 3) differentiating mesothelioma from adenocarcinoma (Morrel, 1995). Western blotting: 5 μg/mL. Sensitivity: 100 mU/mL of human or rat ACE activity (1:20 dilution of human ACE (Catalog number AG 761) or rat ACE (Catalog number AG 782), respectively, which correspond to 100 ng/lane) Optimal working dilutions must be determined by the end user.
