Background Information:
Adipose tissue is highly specialized to play important roles in energy storage, fatty acid metabolism, and glucose homeostasis. Excess of adipose tissue in obesity and lack of adipose tissue in lipodystrophic states are associated with insulin resistance and type-2-diabetes in human and animal studies. Several groups have reported the identification of mouse and human transcript encoding an adipocyte-specific secretory protein originally termed Acrp30 (other names include AdipoQ, apM1, GBP28 and adiponectin). Adiponectin is a protein of 247 amino acids consisting of an N-terminal collagen-like domain and a C-terminal globular domain. The globular domain shares significant homology to the globular domains of type VIII and X collagens, subunits of the complement factor C1q as well as hib27, the latter a protein found in the serum of hibernating mice during non-hibernation. The crystal structures for the globular domain of adiponectin at 2.1 Å resolution reveals significant homology to TNF (tumor necrosis factor) family. Adiponectin circulates as a homotimer or larger complexes of 12-18 subunits. The concentration in serum is approximately 5-10 μg/mL, which corresponds to a maximum concentration of 10-30 nM for the higher order complex. Serum levels of adiponectin are significantly decreased in both obesity and type-2-diabetes, indicating a possible metabolic role or relationship to insulin resistance. In consistence with this, a recent report emphasized a proteolytic fragment (globular domain) of adiponectin to stimulate fatty acid oxidation by muscle.

Application Notes:
Western blot: 1:5,000-1:10,000 using ECL. Reacts with the ~30 kDa adiponectin protein. Suggested blocking and dilution buffer is TBST with 0.05% Tween 20 with 5% skim milk. Suggested incubation time is 1 hour at room temperature. Immunoprecipitation: 1-5 μg per sample in a final reaction volume of 500-1,000 μL. Suggested capture agent is Protein A. ELISA, direct and indirect: 1:5,000-1:10,000. Positive control is mouse plasma. Optimal working dilutions must be determined by the end user.
