It is regulated by Glycogen Synthase Kinase (GSK) 3b, which phosphorylates and targets β-Catenin for ubiquitin-mediated proteolysis. Stabilization of β-Catenin, either by Wnt-mediated inhibition of GSK3b or by mutation (as found in numerous colorectal tumors and melanoma cell lines), results in activation of TCF responsive genes which are thought to play key roles in development and cancer progression. δ-Catenin (NPRAP/Neurojungin), a member of the p120 Catenin subfamily, is a structural protein involved in neuronal development.
