The polyglutamine expansion causes huntingtin to interact abnormally with a number of proteins. However, it is unclear whether, and how, huntingtin-associated proteins are involved in the neurodegeneration in HD. Huntingtin associated protein 1 (HAP-1) is highly expressed in the brain and it interacts with huntingtin, two cytoskeletal proteins (dynactin and pericentriolar autoantigen protein 1), and with a hepatocyte growth factor-regulated tyrosine kinase substrate. The interactions with cytoskeletal proteins and a kinase substrate suggest a role for this protein in vesicular trafficking or organelle transport. Specifically, HAP-1 is involved in intracellular trafficking of epidermal growth factor receptor (EGFR) and is highly expressed in the hypothalamus. View All» Species Reactivity:
