View All» Clone: 8.6 View All» Isotype: IgG1κ View All» Background Information: Interleukin 12 (IL-12) is an interleukin produced by dendritic cells, macrophages and human B-lymphoblastoid cells (NC-37) and is involved in the differentiation of naive T cells into Th1 cells or Th2 cells. IL-12 is a heterodimeric cytokine comprised of p35 and p40 subunits and is considered a T cell stimulating factor. IL-12 stimulates the production of interferon-gamma (IFN-γ) and tumor necrosis factor-alpha (TNF-α) from T and natural killer (NK) cells and reduces IL-4 mediated suppression of IFN-γ. IL-12 can also block the formation of new blood vessels through its induction cascade and is a possible target for anti-cancer drug screening. IL-12 may play a role in autoimmunity by worsening the autoimmune response, due to its key role in induction of Th1 immune responses. Whereas, IL-12 gene knock-out in mice or a treatment of mice with IL-12 specific antibodies seems to ameliorate the diseases. Interleukin 12 is a disulfide-linked heterodimer composed of the 40 kD cytokine receptor like subunit encoded by this gene, and a 35 kD subunit encoded by IL12A. This cytokine is expressed by activated macrophages that serve as an essential inducer of Th1 cells development. This cytokine has been found to be important for sustaining a sufficient number of memory/effector Th1 cells to mediate long-term protection to an intracellular pathogen. Overexpression of this gene was observed in the central nervous system of patients with multiple sclerosis (MS), suggesting a role of this cytokine in the pathogenesis of the disease.
