Background Information:
ARF GTPase-activating protein GIT1 (Git1) is a multidomain protein belonging to the ARF family of small GTPases that is comprised of several protein interacting domains including: three Ankyrin repeats (ANKs), an Arf-Gap domain, a C-terminal GRK-binding domain, and a spa2 homology domain. Git1 is thought to have a role as a signaling pathway coordinator, functioning to regulate adhesion, cytoskeletal organization, and vesicle trafficking. It is also a component of several cytoplasmic complexes that contain ARHGEF6, PAK1, and PXN. In brain receptors, Git1 forms a complex with AMPA receptors, GRIP, and liprin-α, that is crucial to AMPA receptor targeting and the organization of synapses. Git1 expression is observed in a wide variety of tissues, with the most notable expression in the testis.

Sequesters inactive non-tyrosine-phosphorylated paxillin in cytoplasmic complexes.SUBUNIT STRUCTURE: Interacts with SCRIB (By similarity). Interacts with G protein-coupled receptor kinases: ADRBK1/GRK2, PPFIA1 and PPFIA4, with ARHGEF6/alpha-PIX, with ARHGEF7/beta-PIX, with PXN/paxillin and with PTK2/FAK. Component of cytoplasmic complexes, which also contain PXN, ARHGEF6 and PAK1. Interacts with TGFB1I1. SUBCELLULAR LOCATION: Cytoplasm (By similarity).
