Background Information:
Dopamine is a widely distributed neurotransmitter produced by hydroxylation of L-tyrosine, followed by decarboxylation with DOPA decarboxylase. Dopamine exerts its actions by interacting with multiple classes of dopamine receptors that belong to the family of G protein-coupled receptors. Thus far five dopamine receptor subtypes have been reported that are classified into two main families based on their sequence and similarity in signal role. The D1-like dopamine receptors include the dopamine receptors D1 and D5. Binding of dopamine to these receptors leads to the activation of adenylyl cyclase, via a Gs protein, leading to increased cAMP production. The dopamine D2-like receptor group consists of D2, D3, and D4 receptors, which are coupled to Gi/o proteins and act by inhibiting adenylyl cyclase activity. D3 receptor subtype was first cloned and characterized by Sokoloff et al in 1990 (Nature 347, pp 146-151). D3 receptor coupling to various signal transduction mechanisms usually mimic those of D2 receptors, however, their cerebral distribution is very different. Highest expression of D3 receptors is found in ventral striatum in human brain. Dopamine D3 receptor represents a potential target for development of drugs to treat various neuropsychiatric and neurological disorders such as schizophrenia, Parkinson’s disease, drugs of abuse.
