View All» Clone: 1D3 View All» Isotype: IgG1κ View All» Background Information: Protein disulfide-isomerase, also known as PDI, is an abundant component of the ER that catalyzes disulfide bond formation in the biosynthesis of various secretory and cell surface proteins. It is a multifunctional polypeptide identical to the β-subunit of prolyl 4-hydroxylase, a α2β2 tetramer that catalyzes the formation of 4-hydroxyproline in collagen by the hydroxylation of proline residues in peptide linkages. PDI has two thioredoxin-like regions, each containing a Cys-Gly-His-Cys sequence that may represent two independently acting catalytic sites of PDI activity. The C-terminus of PDI has a KDEL sequence, which acts as retention signal for proteins of the ER lumen. The PDI/β-subunit of prolyl-4-hydroxylase is required for its retention in the ER. When present as a tetramer consisting of two alpha and two beta subunits, this enzyme is involved in hydroxylation of prolyl residues in preprocollagen. This enzyme is also a disulfide isomerase containing two thioredoxin domains that catalyze the formation, breakage and rearrangement of disulfide bonds. Other known functions include its ability to act as a chaperone that inhibits aggregation of misfolded proteins in a concentration-dependent manner, its ability to bind thyroid hormone, its role in both the influx and efflux of S-nitrosothiol-bound nitric oxide, and its function as a subunit of the microsomal triglyceride transfer protein complex. At the cell surface, seems to act as a reductase that cleaves disulfide bonds of proteins attached to the cell. May therefore cause structural modifications of exofacial proteins.
