View All» Background Information: Trimethylation of histone H3 on Lys9 (H3K9me3) is one of the most highly studied epigenetic marks. H3K9me3 functions in the repression of euchromatic genes, and in epigenetic control of heterochromatin assembly, most likely by acting as a recognition motif for the binding of chromatin-associated proteins, such as Swi6 or HP1α. The enzymes responsible for H3K9 trimethylation are SUV39H1 and SUV39H2. Suv39H1 is a nuclear protein that acts as a H3K9 specific methyltransferase (HMT). In addition to its role in gene silencing, Suv39H1 is also thought to be involved in heterochromatin organization, chromosome segregation, and mitotic progression, and the epigenetic regulation of telomere length in mammalian cells. Also weakly methylates histone H1 (in vitro). H3 'Lys-9' trimethylation represents a specific tag for epigenetic transcriptional repression by recruiting HP1 (CBX1, CBX3 and/or CBX5) proteins to methylated histones.
