Corneodesmosin forms oligomers and associates homophilically to strengthen the adhesion between corneocytes. Sequential proteolysis of Corneodesmosin diminishes its ability to mediate adhesion, and this is a prerequisite for normal desquamation of the skin. Defects in the synthesis or proteolysis of Corneodesmosin are associated with the development of psoriasis and hypotrichosis of the scalp.
