  View All» Qty/Pk: 96 assays View All» Background Information: Test PrincipleThe UPSTATE colorimetric STAR (Signal Transduction Assay Reaction) ELISA kit is a solid phase sandwich enzyme linked immunosorbent assay that provides a fast, sensitive method to detect specific levels of signaling targets in denatured cell extracts. The ERK 1/2 plate is coated with a specific mouse monoclonal anti-ERK 1/2 capture antibody on the microwells of the 96-well clear plate. Sample lysate or the standard included in the kit are incubated in the microwells allowing ERK 1/2 antigen to be captured in the plate wells. The plate is then washed to remove any non-bound unspecific material. The wells are then incubated with a specific rabbit anti-ERK 1/2 antibody to detect the captured ERK 1/2 on the plate well. The unbound detection antibody is washed away followed by incubation with an HRP-conjugated anti-rabbit antibody. This allows for a sensitive enzymatic detection of the sample. After the addition of TMB substrate and stop solution the absorbance is measured at 450 nm using a plate reader. The entire assay takes less than 5 hours to complete with minimal hands-on time. Many of the reagents are supplied in ready-to use formulations for ease of use. The kit also includes a standard that is run as both a positive control and to develop a standard curve.Erk BackgroundErk (Extracellular signal-Related Kinase) is a family of two, highly homologous proteins denoted as Erk1 (p44, MAPK3) and Erk2 (p42, MAPK1) that both function in the same pathway. The two proteins are often referred to collectively as ERK1/2 or p44/p42 MAP kinase. The Erk pathway is considered the classical, canonical MAPK (Mitogen-Activated Protein Kinase) signaling pathway. It is an evolutionarily conserved pathway that controls and is a critical regulator the growth and survival through the promotion of cell proliferation and the prevention of apoptosis. Erk is involved in the control of many fundamental cellular processes including cell proliferation, survival, differentiation, apoptosis, motility and metabolism. In a normal cell, the signaling pathway is activated by the binding of a growth factor such as EGF, VEGF, PDGF, HGF, and FGF to receptor tyrosine kinases (RTKs). This activates the RTK that starts the cascade through adapter Grb2 that binds to phosphorylated residues of the RTK. SOS (son of sevenless) binds to Grb2 to activate Ras. In its GTP-bound active state, Ras activates the Ser/Thr kinase Raf (MAP Kinase Kinase Kinase). Raf then binds to and activates MEK (MAP Kinase Kinase) by phosphorylating it on the two residue motif. MEK, a dual kinase (a Ser/Thr and a Tyr kinase), phosphorylates and activates MAPK/ERK on the TxY motif (Thr202/Tyr204 and Thr185/Tyr187 for Erk1 and Erk2, respectively). The regulation of a large number of cellular processes are dependent upon the activation state of Erk (p42/p44), so controlling this pathway could have profound effects on various diseases.
