These factors are largely specific for vascular endothelium. The mouse ortholog of human ang4 is called angiopoietin-3. Human Ang4 and mouse Ang3 are strikingly more structurally divergent from each other than are the mouse and human versions of Ang1 and Ang2. Ang3 and 4 are secreted proteins that bind to tyrosine kinase receptor Tie2 and activate it. They are highly expressed in the lung with much lower levels found in other tissues. Ang3 is tethered on the cell surface via heparan sulfate proteoglycans. There is strong evidence that tyrosine kinases are involved in the regulation of tumor progression, cellular growth and differentiation. Ang3 and 4 bind to tyrosine kinase receptor Tie2 and thus their expression is important in several cancers. Overexpression of Ang3 inhibits pulmonary metastasis of Lewis lung carcinoma and TA3 mammary carcinoma cells by inhibiting tumor angiogenesis and promoting apoptosis of the tumor cells. 53% of gastric adenocarcinomas and 79% of colorectal adenocarcinomas show Ang4 expression by immunohistochemistry.
