Specificity:
TACE is a membrane-bound metallo-proteinase-disintegrin. ADAM 17 also processes other cell surface proteins, including TNF receptor, TGF alpha, the L-selectin adhesion molecule, and alpha-cleavage of amyloid protein precursor (APP) (4,5). ADAM 17 mRNA is expressed in a variety of human and murine tissues.

Immunogen:
Raised against a peptide corresponding to amino acids 807-823 of human TACE (1,2). This sequence differs from those of mouse and rat TACE by one amino acid.(3).

Background Information:
ADAM17, also known as TNF-alpha Convertase, or TACE, was first cloned from human epithelial cells. Tumor-necrosis factor alpha is a proinflammatory cytokine and contributes to a variety of inflammatory disease responses and programmed cell death. TNF-alpha is synthesized as a 26 kDa Type II membrane-bound precursor that is cleaved by a convertase to generate secreted 17K mature TNF-alpha. TNF-alpha converting enzyme (TACE) protein was recently purified and the human and mouse TACE cDNAs were cloned by several groups separately (1-3). The ADAM proteins are structurally similar, with a signal sequence, metalloprotease domain (inactive in some ADAMs), disintegrin domain, cystein rich domain, EGF like repeat, type I transmembrane, and a cytoplasmic domain. A member of the metalloproteinase family containing disintegrin like domains (ADAMs), ADAM17 is known to process the TNF alpha trimer from the membrane attached precursor to the soluble form. ADAM17 contains the canonical HExxHxxxxxH zinc metallo-proteinase motif, and has been shown to be proteolytically active, cleaving TNF precursor as well as TNF p75 receptor, myeloid precursor protein, amyloid plaque protein, L selectin and TGF alpha, making ADAM17 a "sheddase". Reports have shown that ADAM17 expression in human breast tumor samples is an indicator of poor prognosis.

Application Notes:
Western blot: 1:500 - 1:2,000 dilution from a previous lot was used. HeLa or Jurkat whole cell lysate can be used as a positive control and 80 to 130 kDa bands can be detected which may represent mature protein, precursor, and glycosylated TACE.Immunocytochemistry: Optimal working dilutions must be determined by end user.

Entrez Gene Summary:
This gene encodes a member of the ADAM (a disintegrin and metalloprotease domain) family. Members of this family are membrane-anchored proteins structurally related to snake venom disintegrins, and have been implicated in a variety of biologic processes involving cell-cell and cell-matrix interactions, including fertilization, muscle development, and neurogenesis. The protein encoded by this gene functions as a tumor necrosis factor-alpha converting enzyme; binds mitotic arrest deficient 2 protein; and also plays a prominent role in the activation of the Notch signaling pathway.

UniProt Summary:
FUNCTION: SwissProt: P78536 # Cleaves the membrane-bound precursor of TNF-alpha to its mature soluble form. Responsible for the proteolytic release of several other cell-surface proteins, including p75 TNF-receptor, interleukin 1 receptor type II, p55 TNF-receptor, transforming growth factor-alpha, L-selectin, growth hormone receptor and the amyloid precursor protein. Also involved in the activation of Notch pathway (By similarity).COFACTOR: Binds 1 zinc ion per subunit.SIZE: 824 amino acids; 93021 Da SUBUNIT: Interacts with MAD2L1.SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.TISSUE SPECIFICITY: Ubiquitously expressed. Expressed at highest levels in adult heart, placenta, skeletal muscle, pancreas, spleen, thymus, prostate, testes, ovary and small intestine, and in fetal brain, lung, liver and kidney.DOMAIN: SwissProt: P78536 Must be membrane anchored to cleave the different substrates. The cytoplasmic domain is not required for the this activity. Only the catalytic domain is essential to shed TNF and p75 TNFR (By similarity). & The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion, thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.PTM: The precursor is cleaved by a furin endopeptidase (By similarity). & Phosphorylated. Stimulation by growth factor or phorbol 12- myristate 13-acetate induces phosphorylation of Ser-819 but decreases phosphorylation of Ser-791.SIMILARITY: Contains 1 disintegrin domain. & Contains 1 peptidase M12B domain.
