The antibody is made with a synthetic peptide based on the carboxyterminal end of active MMP-23, using the human sequence. The antibody binds to MMP-23, but does not cross react with the other MMP family members (MMP-1, MMP-2, MMP-3, MMP-9, etc.).Current nomenclature gives the titles MMP-21 and MMP-22 to the Chicken and Xenopus MMPs. Sequences for MMP-23 were lodged under the names MMP-21, MMP-22, MIFR-1 and MIFR-2; all of which are variants of MMP-23. MMP-23, first described in tumor cells, shares much of the same domain structure of the classical MMPs, but has a unique cysteine array. Also, the cysteine switch found in the classical MMPs is lacking or has a different sequence. MMP-23 is most similar to MMP-11, but there are significant differences between the two. MMP-23 has a furin cleavage site, and is thought to be localized within cells. MMP-23 has been reported as several splice variants with different truncations at the carboxy terminus. The zymogen for MMP-23 is about 58 Kd, and is quickly activated to the 45 Kd form, and this breaks down to a cascade of active forms. AB19095 Anti-MMP-23 works well for Western Blotting applications. AB19095 will also bind to the non-reduced protein, and thus has application for immunoprecipitation, immunohistochemistry, and ELISA. MMP-23 levels in quiescent cells and tissues are minimal, and mitogen stimulation or protein concentration is often needed to visualize the bands. In addition, cell types differ greatly in the quantity of MMP-23 produced. MMP-23 is thought to be localized within cells, but may also be seen in conditioned media from cells in culture.
