View All» Background Information: BCL2/adenovirus E1B 19 kDa protein-interacting protein 3 (BNIP3) is a member of the NIP3 family, a singular group of death-inducing, mitochondrial proteins. BNIP3 is comprised of several domains of interest; a highly conserved domain (CD), a C-terminal transmembrane (TM) domain, and a BH3 domain. Strongly upregulated by hypoxia, BNIP3 induces a caspase-independent cell death characterized by mitochondrial damage. Overexpression has been observed in the brain after stroke and in the adult heart following myocardial infarction. Recent studies have shown BNIP3 to be involved in cell growth inhibition through suppression of the mTOR pathway and that, when localized to the nucleus, it has the paradoxical capability of repressing apoptosis in tumors by binding the AIF expression gene.
