The antiserum has been tested on tissue sections from the rat central nervous system (CNS) using immunofluorescence histochemistry. The antiserum preferentially labels cell bodies in the hippocampal formation, cerebral cortex, thalamic nuclei, Purkinje cells of the cerebellum, motoneurons in the brainstem and spinal cord. The staining pattern for the immunoreactive cell bodies obtained with the nicastrin antiserum corresponds to the pattern described using antibodies to presenilin-1. Direct double-labeling immunofluorescence histochemistry shows co-localization of nicastrin with presenilin-1-immunoreactivity in a large number of neurons. The antiserum has so far only been tested on rat tissue. Due to sequence homology, the antiserum is predicted also to work on human tissue. The antiserum has so far not been tested using Western blotting. SPECIES REACTIVITIES: It is expected that the antibody will also react with mouse and human based on sequence homology (amino acids 691-709). Other species have not yet been tested.
