The 21 kDa active form is not recognized by the antibody. This antibody will also bind to the non-reduced protein, and thus has application in immunoprecipitation, immunohistochemistry, and ELISA. Stromelysin levels in quiescent cell cultures and tissues is minimal. Stimulation of synthesis or concentration of the protein from culture medium is often needed to visualize blotted bands. Cell types differ greatly in the quantity of Stromelysin produced. A recommended starting concentration for Western Blots is 1:1,000 when using colorimetric substrates such BCIP/NBT, and 1:5,000 for chemiluminescent substrates.
