View All» Background Information: MLL is a histone methyltransferase and chromatin modifier that plays a role in hematopoiesis. This protein is implicated in HOX gene regulation in early embryogenesis and in the regulation of non-HOX genes p27 (Kip1) and p18 (lnk4C). It is part of a catalytic subunit of the MLL1/MLL complex, which mediates the acetylation of Lys-16 of histone H4 (H4K16ac) and the methylation of Lys-4 of histone H3 (H3K4me). It is reported that MLL interacts directly with the polymerase associated factor complex (PAFc) through sequences flanking the CxxC domain. Subsequently, PAFc interacts with RNA polymerase II and thereby stimulates posttranslational histone modifications. Taspase 1, catalyzes MLL into MLL-C and MLL-N, both of which reassociate and later reassemble into different multiprotein complexes that regulate the transcription of specific target genes, including many of the HOX genes. The encoded protein contains multiple conserved functional domains. One of these domains, the SET domain, is responsible for its histone H3 lysine 4 (H3K4) methyltransferase activity which mediates chromatin modifications associated with epigenetic transcriptional activation. This protein is processed by the enzyme Taspase 1 into two fragments, MLL-C and MLL-N. These fragments reassociate and further assemble into different multiprotein complexes that regulate the transcription of specific target genes, including many of the HOX genes.
