View All» Background Information: Pleckstrin (PLEK) is the major substrate target for phosphorylation by protein kinase C (PKC) in platelets. A short stretch of three amino acids (Ser113, Thr114 and Ser117), towards the NH2-terminus of PLEK is targeted for phosphorylation by PKC. One of the earliest events following platelet stimulation is the phosphorylation of the 40 kDa protein pleckstrin. Most platelet stimuli result in PLEK phosphorylation. These include alpha-thrombin, collagen, phorbo 12-myristate 13-acetate (PMA), thrombin receptor activating peptide, serotonin and thromboxane A2. Furthermore, the phosphorylation of plek has been shown to correlate with platelet secretion. PLEK-null platelets from a knockout mouse have been shown to be defective in granule secretion, aggregation and actin polymerization.
