View All» Background Information: Phosphodiesterase 4A4 (PDE4A4) is a member of the PDE4 family of cyclic AMP degrading phosphodiesterases. PDE4A4 seems to have a biphasic Mg(2+) dose-response in catalyzing cyclic AMP hydrolysis. The rodent orthologue of PDE4A4, which is called PDE4A5, has an identical N-terminal region to human PDE4A4 and is detected equally effectively and specifically by this antiserum. PDE4A4 is a long PDE4 isoform whose regulatory UCR1 module can be phosphorylated by PKA, leading to activation. Its UCR1 module can also be phosphorylated by MK2, the downstream kinase of p38 MAPK, and this action attenuates the degree of activation elicited by PKA phosphorylation. PDE4A4 is able to be sequestered into distinct signaling complexes involving either the p75 neurotrophin receptor (p75NTR), beta-arrestin, various SH3 domain - containing proteins and DISC1.
