View All» Background Information: Ubiquitin-specific-processing protease 33 (USP33) is a widely expressed, deubiquitinating enzyme first identified as a von Hippel-Lindau tumor suppressor (VHL) protein-interacting deubiquitinating enzyme (hVDU1) and is a member of the peptidase C19 family. USP33 is involved in a variety of processes including beta-2 adrenergic receptor/ADRB2 recycling and cell migration. Known to bind Robo1 receptor, USP33 is a required component of the Slit signaling pathway responsiveness in breast cancer cells. Slit redistributes Robo1 to the plasma membrane from intracellular compartments in a manner dependent on the presence UP33, which produces an inhibitory effect on breast cancer cell directional migration. USP33 has been shown to bind to and deubiquitinate D2 as a crucial response to cold exposure, helping to decrease D2 degradation, and acclimatize brown adipocytes. Involved in cell migration via its interaction with intracellular domain of ROBO1, leading to regulate the Slit signaling. Plays a role in commissural axon guidance cross the ventral midline of the neural tube in a Slit-dependent manner, possibly by mediating the deubiquitination of ROBO1. Acts as a regulator of G-protein coupled receptor (GPCR) signaling by mediating the deubiquitination of beta-arrestins (ARRB1 and ARRB2) and beta-2 adrenergic receptor (ADRB2). Plays a central role in ADRB2 recycling and resensitization after prolonged agonist stimulation by constitutively binding ADRB2, mediating deubiquitination of ADRB2 and inhibiting lysosomal trafficking of ADRB2. Upon dissociation, it is probably transferred to the translocated beta-arrestins, leading to beta-arrestins deubiquitination and disengagement from ADRB2.
