GAK promotes microtubule growth during mitotic spindle assembly and is required for centrosome maturation, chromosome congression, and mitotic progression. It also regulates the assembly and intracellular trafficking of clathrin-coated vesicles. GAK contains a Ser/Thr protein kinase domain (aa 40-314), a phosphatase tensin/auxilin domain (aa 399-566), a PTEN C2 domain (aa 570-708), and a J domain (aa 1247-1311).
