人BMP-7基因定位于2号染色体,其cDNA编码区有1293个碱基,含有1个开放读码框,编码多肽产物由431个氨基酸残基组成,起始序列为一段由29个氨基酸残基组成信号肽,中间第29-292个氨基酸残基称为前肽,第293-431个氨基酸残基称为成熟肽。该前体蛋白合成分泌后,在体内被剪切加工成汉139个氨基酸残基成熟肽,每个成熟有活性BMP-7都是由二硫键连接成同源或异源二聚体。BMP-7具有多方面生物学效应,包括在胚胎发生3个阶段发挥作用,在肾脏形态发生中也起到重要作用,另外BMP-7还具有诱导成骨作用,能促成骨细胞增殖分化,可诱导软骨细胞分化,在骨代谢中发挥重要作用。

人骨形态发生蛋白-7,CHO表达重组蛋白是在CHO中国仓鼠卵巢细胞中重组表达的。其序列结构为“N末端--人BMP-2蛋白(Met 1- Arg 282)--人BMP-7蛋白(Ser 293 - Arg 431)--C末端”。表达人BMP-2的信号肽和1-282个氨基酸序列与人BMP-7成熟肽293-431个氨基酸序列融合表达于CHO细胞。最后成熟的BMP-7蛋白是通过蛋白酶水解掉包含信号肽和前肽的139个氨基酸而形成。糖基化增大了人骨形态发生蛋白-7分子量,使非还原状态下在SDS-PAGE电泳时条带位于25-40kDa。该人骨形态发生蛋白-7,CHO表达重组蛋白最终由标准的色谱技术纯化出。

溶解:提供的人骨形态发生蛋白-7,CHO表达(BMP 7 CHO)产品为冻干粉末;该冻干粉末是从含1mg/ml人骨形态发生蛋白-7,CHO表达(BMP 7 CHO)的1%蔗糖,1.2%甘露醇,20mM 氨基乙酸和0.05% Tween 20 (pH 4)溶液中灭菌并冻干产生的。我们建议加1ml无菌且不含内毒素的水以配制工作储存液。

保存:人骨形态发生蛋白-7,CHO表达(BMP 7 CHO)冻干粉末在室温可保存3周,或置于-18℃冻存用于长期保存。重新溶解后可以在4℃冷藏保存至少2-7天以上;如果想延长保存时间,建议分装(加载体蛋白如0.1% HSA或 BSA)后置于-18℃冻存。请避免反复冻融。

Introduction (介绍)

The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development. In addition, the fact that this BMP is closely related to BMP5 and BMP7 has lead to speculation of possible bone inductive activity.

Description (描述)

N-TERMINAL---Human BMP-2 (Met 1 – Arg 282) Human BMP-7 (Ser 293 – Arg 431)---C-TERMINAL. The DNA sequence encoding the human BMP-2 signal peptide and propeptide (1~282 amino acid) fused to the human rhBMP-7 mature chain (293~431 amino acid) was expressed in a Chinese hamster ovary cell line. The mature recombinant BMP-7 generated by the proteolytic removal of the signal peptide and propetide contains 139 amino acid residues. The glycosylation of BMP-7 increases the molecular mass and the glycosylated proteins migrate as 25 ~ 40 kDa in SDS-PAGE under non-reducing conditions.

BMP-7 is purified by proprietary chromatographic techniques.

Formulation

BMP-7 was lyophilized from a concentrated (1mg/ml) sterile solution containing 1% sucrose, 1.2% mannitol, 20mM glycine, and 0.05% tween 20 pH-4.

Solubility (溶解性)

It is recommended to reconstitute the lyophilized Bone Morphogenetic Protein-7 in 1ml sterile/ endotoxin free water.

Stability (稳定性)

Lyophilized BMP-7 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution BMP 7 Human should be stored at 4°C between 2-7 days and for future use below -18°C.

For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).

Please prevent freeze-thaw cycles.

Purity (纯度)

Greater than 97.0% as determined by:

(a) Analysis by RP-HPLC.

(b) Analysis by SDS-PAGE.

Biological Activity (生物活性)

Measured in alkaline phosphatase activity assay using MC3T3-E1 cells. The ED50 for this effect is <100ng/ml, corresponding to a Specific Activity of 10,000IU/mg.
